Cell Cycle-Associated Appearance of Glycoproteins Containing Terminal N-Acetylglucosamine in a WI-38 Human Cell Line

Abstract
Abstract:
@EN Glycocomponets of mammalian cells have been implicated in stage-specific associated changes during the cell cycle. The study was performed to examine the appearance of N-acetylglucosamine(GlcNAc)-containing glycoproteins in mammalian cells.
WI-38
normal embryonic human lung fibroblast cells were cultured in DMEM containing fetal calf serum. Cells were harvested after appropriate treatment to recover synchronized cells at a specific point in the cell cycle, Glycoproteins containing GlcNAc
were
detected using peroxidase-conjugated wheat germ agglutinin(P-WGA) and the GlcNAc terminal were labeled with [3H]-galactose. Isolated mouse liver nuclei cotained various glycoproteins, and synchronized WI-38 cells in G0 and M phases showed
different
patterns of glycoproteins. The appearance of GlcNAC-containing glycoproteins may be associated with stage-specific cellular activity occurring in the normal cell cycle.
